PPT Tubes / Plasma Preparation Tubes (Molecular NAT Testing)

XINLE PPT Tubes (Plasma Preparation Tubes) are sterile, evacuated blood collection tubes pre-loaded with spray-dried K2EDTA anticoagulant and a specialized inert separation gel. Designed for high-sensitivity molecular diagnostics and viral Nucleic Acid Testing (NAT), these CE-certified PET tubes form a stable physical barrier upon centrifugation, allowing undiluted plasma to be processed, transported, and stored directly in the original tube at ≤ -20°C for up to 5 years or at -80°C for biobanking.

Product Overview

Manufactured under strict ISO 13485 quality management systems and bearing the CE mark, XINLE PPT Tubes (Plasma Preparation Tubes) provide an integrated, closed-system workflow for molecular diagnostics, blood banking, and infectious disease screening.

In conventional molecular workflows, transferring separated plasma into secondary aliquoting tubes increases hands-on time, sample cross-contamination, and exposure to bloodborne pathogens. Our PPT tubes integrate whole blood collection, barrier gel separation, transport, and long-term in-tube cryopreservation. Featuring an ultra-clean, pyrogen-free environment with endotoxin levels controlled at ≤0.005 EU/mL and certified DNase/RNase-free, PPT tubes safeguard target viral DNA and RNA from enzymatic degradation and hemoglobin interference.

Key Features & Biological Advantages

  • Integrated In-Tube Cryopreservation (≤ -20°C to -80°C): Separated plasma remains sealed in the primary tube above the inert gel barrier, supporting long-term frozen storage at ≤ -20°C for up to 5 years and full compatibility with -80°C ultra-low temperature biobanking.
  • Inert Gel Physical Barrier: Centrifugation causes the thixotropic gel to form an impermeable physical barrier between the plasma and blood cells, preventing cellular leakage, red blood cell lysis, and free hemoglobin interference.
  • Spray-Dried K2EDTA (Undiluted Plasma): Precision spray-coated K2EDTA anticoagulant eliminates liquid dilution effects, yielding undiluted plasma specimens that enhance the analytical sensitivity and reproducibility of PCR/qPCR assays.
  • Ultra-Low Endotoxin & Pyrogen-Free (≤0.005 EU/mL): Strict cleanroom bio-burden control eliminates background pyrogenic and endotoxin interference.
  • Certified DNase-Free & RNase-Free: Batch-tested on dedicated nuclease detection platforms to ensure minimal endogenous enzyme activity, preserving low-copy viral RNA and DNA targets.
  • Taq Polymerase Compatibility: High-purity K2EDTA formulation exerts zero inhibitory effects on downstream Taq polymerase enzymatic activity during amplification.
  • Reduced Manual Handling: Eliminates manual plasma pipetting/aliquoting into secondary cryovials, drastically cutting consumable costs, labor time, and biohazard exposure risks.

Clinical & Molecular Diagnostic Applications

XINLE PPT Tubes are designed for high-quality plasma preparation for infectious disease Nucleic Acid Amplification Testing (NAAT):

  • Hepatitis B Virus (HBV DNA) Testing: Quantitative and qualitative HBV viral load testing for chronic infection monitoring and blood screening.
  • Hepatitis C Virus (HCV RNA) Testing: High-fidelity viral RNA amplification for active infection confirmation and direct-acting antiviral (DAA) therapy response tracking.
  • Human Immunodeficiency Virus (HIV RNA) Testing: Viral load quantification for early HIV diagnosis, antiretroviral therapy (ART) monitoring, and blood bank safety screening.
  • Treponema Pallidum (Syphilis) DNA Testing: Molecular nucleic acid amplification testing assisting in definitive syphilis infection diagnosis.
  • Blood Donor Screening & Biobanking: Routine closed-system NAT screening in blood transfusion centers and long-term cohort specimen storage.

Operation Guide: Centrifugation & Storage Workflow

Follow this standardized operational sequence to prepare and cryopreserve plasma:

  1. Blood Draw: Perform routine venipuncture using a standard evacuated blood collection needle. Draw blood to the nominal fill volume (5 mL, 8 mL, or 8.5 mL).
  2. Inversion Mixing: Gently invert the tube 8 to 10 times immediately after blood collection to mix the whole blood thoroughly with the spray-dried K2EDTA.
  3. Centrifugation: Centrifuge the tube in a swinging-bucket centrifuge according to standard laboratory molecular protocol. The inert gel migrates to form an impermeable barrier above the erythrocyte and leukocyte pellet.
  4. Direct Storage or Transport: Store or ship the separated plasma directly in the original sealed tube. For long-term biobanking, place the intact tube directly into -20°C or -80°C freezers without aliquoting.

Frequently Asked Questions (FAQ)

A: Pyrogens and bacterial endotoxins can interfere with sensitive molecular enzymatic assays and trigger false background reactions in high-sensitivity laboratory applications. XINLE PPT Tubes undergo strict quality control of raw materials, cleanroom environments, and automated manufacturing processes to maintain an ultra-low endotoxin threshold (≤0.005 EU/mL), ensuring dependable sample integrity for advanced nucleic acid testing.

A: A PPT Tube (Plasma Preparation Tube) is a specialized evacuated blood collection tube designed for integrated plasma collection, preparation, transportation, and long-term storage in molecular diagnostic workflows. It contains spray-dried K2EDTA anticoagulant and an inert separation gel, enabling rapid plasma isolation and contamination-free preservation for downstream viral load and nucleic acid testing (NAT).

A: PPT Tubes provide a certified DNase-free and RNase-free internal environment that actively prevents the enzymatic degradation of fragile viral DNA and RNA molecules. Furthermore, each production batch undergoes dedicated nuclease release testing to guarantee maximum nucleic acid stability prior to downstream PCR amplification.

A: Yes. We provide comprehensive OEM and custom manufacturing solutions, including custom tube dimensions, specific draw volumes, customized additive formulations, private label branding, and tailored packaging configurations according to your laboratory’s clinical and automation requirements.
